Iron chelators as a therapeutic option for Alzheimer’s disease—A mini-review

Neurodegenerative disorders, particularly Alzheimer’s disease (AD), remain a great challenge regarding the finding of effective treatment, one main reason being incomplete understanding their etiology. With many intensely debated hypotheses, newer approach based on impact iron imbalance in sustaining neurodegeneration central nervous system becomes increasingly popular. Altered homeostasis leads to increased accumulation specific brain areas, explaining clinical picture AD patients. Moreover, growing evidence sustains significant metabolism relationship other pathological processes encountered AD-affected brain, such as amyloidogenic pathway, chronic inflammation, or oxidative stress. In this context, mini-review aims summarize novel data from continuously expanding literature topic didactic manner. Thus, first part, authors briefly highlight most relevant aspects related absorption, transport, regulation, and elimination at cerebral level, focusing role blood-brain barrier concept ferroptosis. Subsequently, currently available chelation therapies are discussed, including an overview trials topic. final latest results vitro vivo studies, new research directions suggested enhance development antidementia therapies.
